To understand the gravity-response in plants regulating auxin distribution, we investigate cucumber gravimorphogenesis and Arabidopsis root gravitropism. When cucumber (Cucumis sativus L.) seeds germinate in a horizontal position, a protuberance called a peg forms on the lower side of the transition zone. It has been suggested that activities of auxin efflux carriers in the upper side of the transition zone is facilitated by gravity-response. To understand the mechanism of decrease of auxin in the upper side of the transition zone in response to gravity, we are investigating the accumulation of auxin efflux carriers in the transition zone of cucumber seedlings. In addition, we are attempting to isolate novel root gravitropism mutants of Arabidopsis sbecause a mutant that is defective in regulating the relocalization of auxin efflux carrier in root columella cells by gravistimulation has not been isolated. We have screened approximately 100,000 EMS-mutagenized M2 seedlings, and obtained 44 lines that would be defective in root gravitropism.
