抄録 | Active transport of ions is a ubiquitous process in biological membranes. To understand its fundamental mechanism, a retinal-containing protein, bacteriorhodopsin (bR), that works as a light-driven proton pump in the plasma membrane of halobacteria has been studied. How the proton pumping cycle of bR is affected by environmental factors has been elucidated including the protein flexibility, and recently a hypothesis that a sliding motion of transmembrane helical segments in the protein is involved in a principal step of proton transport is proposed with a view to studying such a dynamic structure at an atomic resolution, it has been challenged to get a high-quality crystal of bR. イオンの能動輸送は生物膜においては極く普通のプロセスである。その基礎メカニズムを理解するため、ハロバクテリアのプラズマ膜において光により駆動される陽子ポンプの働きをする網膜に含まれる蛋白質、すなわちバクテリオロドプシン(bR)に関する研究を行った。bRの陽子ポンピングサイクルが蛋白質の柔軟性を含む環境因子によりどのような影響を受けるかを明らかにするとゝもに、陽子輸送の主要なステップには、蛋白質中のトランス膜の螺旋セグメントの滑り運動が含まれているという仮説を最近になって提出した。このような動的構造を原子レベルの分解能で研究するという観点から、高品位のbR結晶を得るべく試みているところである。 |
