抄録 | A polyhedral structure in a novel crystal of the membrane protein bacteriorhodopsin was discovered. Atomic force microscope images of the crystal surface indicate that bacteriorhodopsin polyhedra with a diameter of 50 nm are hexagonally close-packed. The polyhedra contain native lipids and, at low ionic strength, they fuse with one another to form tubular or doughnut-like structures. Large-scale morphological changes are also observed in other biological membranes. On the basis of these observations, the concept of membrane protein polymorphism, which may be a key to better understanding of the dynamic structure of biological membranes is introduced. 膜蛋白質バクテリオロドプシンの新しい結晶における多面体構造を見いだした。結晶表面の原子力顕微鏡画像は、直径50mmのバクテリオロドプシン多面体が6方晶系的に密集していることを示している。多面体は固有の脂質を含み、低イオン強度で互いに融合し、管状またはドーナッツ状構造を形成する。その他の生体膜では大規模な形態的変化が見られた。これらの観察にもとずいて、膜蛋白質の多形性の概念を紹介するが、これは生体膜の動的構造の理解にとって優れたキーとなる。 |
