抄録The effects of ML-9, a myosin light chain kinase inhibitor, on Ca(2+) mobilization and contraction induced by Methacholine (MCh) in guinea pig tracheal smooth muscle was examined. Isometric tension and intracellular Ca(2+) concentration ((Ca(2+))(sub i)) assessed by Ca(2+) indicator fura-2 were measured simultaneously. MCh (1 micro M) induced a sustained increase in (Ca(2+))(sub i) and in tension. ML-9 (100 micro M) reduced both (Ca(2+))(sub i) and contraction by MCh. There was an apparent discrepancy in the time course of reduction of between (Ca(2+))(sub i) and tension. These results suggest that ML-9 reduced MCh-induced increases in (Ca(2+))(sub i) not by MLCK inhibition but by direct effects on Ca(2+) entry in guinea pig tracheal smooth muscle.
